(A) Cells were incubated with the indicated RNAi oligos for 40 hrs, followed by a medium change to Assay Medium. Cells were then stimulated with 400 nM 17-AAG for 6 hours before the beta-lactamase assay was performed as described in Methods. (B) Cells were incubated with the indicated RNAi oligos for 40 hrs, followed by heat shock at 42°C for 1 hour and a 5-hour recovery at 37° C before the beta-lactamase assay was performed as described in Methods (n=3 for each data point).
